What is Glucotize™?
Glucotize™ is a-lipoic acid, a powerful antioxidant, delivered in a proprietary controlled release formulation developed by Medical Research Institute (MRI; San Bruno, CA). The controlled release formulation was developed by MRI to significantly extend the amount of time a-lipoic acid is present in human plasma. This enables a-lipoic acid to achieve its maximum physiological benefits.

What is a-lipoic acid?
a-Lipoic acid is a short-chain fatty acid that contains two sulfur atoms. It is a natural compound that is produced in small quantities in all human cells, and functions as a component of the pyruvate dehydrogenase and a-ketoglutarate dehydrogenase mitochondrial enzyme complexes. These enzymes are extremely important in oxidative glucose metabolism. In other words, these enzymes are necessary to convert glucose into cellular energy (ATP).

In standardized doses (600-900 mg), a-lipoic acid and its reduced form, dihydrolipoic acid (DHLA), function as extremely potent and effective antioxidants. There are numerous scientific studies that have shown that a-lipoic acid is able to restore and recycle the endogenous network of antioxidants including Vitamins C and E, and glutathione. Thus, a-lipoic acid functions in concert with other antioxidants. Other work has indicated that a-lipoic acid possesses metal-chelating activity, reactive oxygen scavenging activity, and plays a role in the repair of proteins, DNA, and lipids.

Is thioctic acid the same as a-lipoic acid?
Yes, thioctic acid is the term originally given to a-lipoic acid because the compound contains sulfur (thio). Since this compound is a fatty acid (lipid) and therefore soluble in fat (lipid), it is more commonly referred to as a-lipoic acid.

There are many a-lipoic acid products. What is the advantage of MRI’s Glucotize™?
Virtually all commercially available a-lipoic acid supplements can be referred to as immediate release products, whether supplied in the form of a tablet or capsule. The term immediate release means that once a-lipoic acid passes through the stomach and into the upper gastrointestinal tract, it is immediately begins to be absorbed into the plasma. In fact, within 20-30 from the time that you take immediate release a-lipoic acid, it reaches its maximum concentration in the plasma. Virtually all of a-lipoic acid taken in the immediate release form is cleared (removed) from plasma within ninety minutes. The end result is that the a-lipoic acid is not in the body long enough to provide its maximum physiological benefits, including glucose lowering or relieving the conditions associated with unhealthy blood sugar levels.

The primary advantage of Glucotize™ is it overcomes the significant limitation of all immediate release a-lipoic acid products, i.e. an abbreviated plasma half-life. Glucotize™ delivers a-lipoic acid in a controlled release fashion. This formulation significantly extends the amount of time a-lipoic acid is present in human plasma (referred to as plasma half-life). This enables a-lipoic acid to achieve its maximum physiological benefits, above and beyond what is possible with other a-lipoic acid products.

What is the plasma half-life of a-lipoic acid in immediate release products compared to Glucotize™?
Immediate release forms of a-lipoic acid possess a plasma half-life of approximately 30 minutes, while MRI’s Glucotize™ has a half-life of over four times longer. The other products typically use an older technology, called enteric coating. This process coats the tablets to prevent a quick delivery; however, it does not regulate the delivery of the product. MRI has developed a proprietary formula using state-of-the-art technology that is commonly used in the pharmaceutical industry. The end result is that MRI’s Glucotize™ delivers a-lipoic acid in a controlled, regulated rate over several hours.

If "immediate release" a-lipoic acid products possess limited usefulness, why do so many other companies sell the product?
MRI has invested over $4,000,000 to develop and optimize the delivery technology employed in Glucotize™. This formulation extends the physiological benefits of a-lipoic acid. This substantial investment is a major hurdle for most nutraceutical and dietary supplement manufacturers. Thus, this is the reason the consumer is able to find so much of the less expensive and efficacious products on the shelf today.

Why is Glucotize™ more expensive than other a-lipoic acid products?
Glucotize™ was developed over a two-year period. It is the result of a systematic evaluation of many different controlled release formulations. This process was tedious, time-consuming, and expensive. However, it was necessary to identify a formulation that offers the most efficacious preparation of a-lipoic acid commercially available.

Unfortunately, many nutraceutical and dietary supplement companies manufacture and market products lacking any peer-reviewed clinical (scientific) data to support their claims. In contrast, MRI invests a considerable percentage of its operating budget in support of clinical research. For example, MRI has just completed a pharmacokinetic study designed to compare the controlled release profile of Glucotizeä with that of immediate release a-lipoic acid. This study evaluated Glucotizeä and a major brand of immediate release a-lipoic acid in normal volunteers over a twenty-four hour period. The results of this study will be available very soon. Briefly, this study found that the immediate release form of a-lipoic acid possess a plasma half-life of approximately 30 minutes, while MRI’s Glucotize™ has a half-life of approximately 4 times longer.

MRI is also sponsoring a second clinical study designed to evaluate the safety, tolerability, and efficacy of Glucotize™ in people with impaired blood sugar metabolism. This study is just about complete; the results will be available shortly. The data obtained thus far indicate that Glucotize™ is very well tolerated, and has not produced any significant side effects. Of equal importance are the observations that show Glucotize™ is able to significantly lower plasma fructosamine levels. Fructosamine is glycated albumin and provides an approximation of glucose control over a three-week period. This is an important clinical finding, and has not been achieved previously with any other a-lipoic acid product.

Other factors that contribute to the cost of Glucotize™ are associated with the costs of manufacturing. Unlike other a-lipoic acid products, Glucotize™ is manufactured using state-of-the-art technology frequently employed in the pharmaceutical industry for controlled release formulations. In addition, Glucotize™ is manufactured in accordance with cGMPs (Good Manufacturing Practice) environment. The MRI emphasis on the highest quality manufacturing techniques and conditions ensures that Glucotize™ is manufactured under the strictest standards ensuring maximum batch-to-batch reproducibility, safety, and efficacy. Thus, while the cost of Glucotize™ may be slightly higher than other a-lipoic acid products, MRI guarantees that you are being provided with the best and most efficacious form of a-lipoic acid commercially available.

Are there clinical data indicating that a-lipoic acid is present in human plasma longer when administered as Glucotize™?
Yes. As described above, MRI recently sponsored a clinical study to investigate this important question. This study employed normal volunteers, who were given 600 mg of either Glucotize™ or immediate release a-lipoic acid. This study was a crossover design, which means that all subjects eventually received both Glucotize™ and immediate release a-lipoic acid. The amount of lipoic acid in their plasma was determined over a twenty-four hour period using a validated bioanalytical assay. The results indicate that Glucotize™ is detected in human plasma for up to four times longer than immediate release a-lipoic acid.

Why should I take Glucotize™?
Glucotize has many beneficial health effects. Principal among these are the restoration and recycling of the endogenous antioxidant network including Vitamins C and E, ability to increase tissue glutathione levels, and the enhancement of mitochondrial function. Due to the negative impact caused excessive free radical accumulation and the importance of the role that antioxidants play in neutralizing free radicals, a-lipoic acid delivered in Glucotize™ plays a major preventative role. In addition, there are an increasing number of studies in animals showing the benefits of a-lipoic acid in delaying the aging process. Glucotize™ is an important addition to the dietary supplement regimen for any healthy individual.

Glucotize is especially beneficial for people with unhealthy blood sugar levels. Upon use, you can expect a reduction in fasting plasma glucose levels with a dosage of three to four tablets a day. Are there any clinical data to support the safety and/or efficacy of Glucotize™?
Yes. MRI is in the final stages of completing a clinical trial with 17 patients using Glucotize™ for twelve weeks. In these subjects, Glucotize™ was tolerated very well. No patients left the study because of Glucotize™-related adverse events or side effects. There were no changes in liver enzymes, or in any other surrogate marker of safety or tolerability. Importantly, this study has shown promising results for lowering plasma fructosamine levels as well as some other critical parameters. Fructosamine is glycated albumin, and serves as a marker of glucose control over a three-week period.

What is the recommended dose of Glucotize™?
It is suggested to take one tablet before breakfast and one tablet before dinner. If additional support is needed to maintain healthy blood sugar levels, try taking two tablets before breakfast and one tablet before dinner. In all cases, it is recommended to take Glucotize 30 – 60 minutes before eating, as some foods might interfere with the absorption of a-lipoic acid.

Is Glucotize™ safe?
Are there any side effects and are there any reasons why it should not be taken? Are there any known interactions of Glucotize™ with other medications?

In normal individuals and those with unhealthy blood sugar levels, Glucotize™ has demonstrated a very high level of safety and tolerability when used at the recommended doses. As with any modification in disease management, you should consult with your personal physician or pharmacist before initiating the change. Also, women who are pregnant or lactating should consult their personal physicians before using this product.

Is a-lipoic acid present in any food? Can any benefit of a-lipoic acid be obtained from food sources, and is it as effective as Glucotize™?
Spinach, potatoes and red meat are the richest natural sources of a-lipoic acid. Spinach provides the highest relative amount of a-lipoic acid among any food tested, containing approximately 3 milligrams (mg) of a-lipoic acid per gram of dry weight. However, to receive 300 mg of a-lipoic acid from spinach you would have to eat 100 grams of dried spinach (which corresponds to several kilograms of wet weight). Clearly, this is an impossible task. However, even if you could afford to buy and eat large quantities of spinach everyday, you would not achieve the controlled release properties of Glucotize™, which delivers, over several hours, a-lipoic acid to your blood plasma.

Does Glucotize help promote nerve growth or regeneration?
Oxidative stress is markedly increased in individuals suffering from a variety of conditions. Increased oxidative stress compromises the functions of nerve cells, inhibits their ability to respond to endogenous stimulators of nerve growth such as nerve growth factor (NGF), and accelerates the rate of nerve cell death. In addition, oxidative stress decreases the blood flow supplying these cells, depriving them of essential nutrients. In its antioxidant role, the a-lipoic acid delivered in Glucotize™, promotes the neutralization of free radicals responsible for oxidative stress.
